Defining Clinically Relevant Protein-Glycan Interactions

Glycans are complex biomolecules made from sugars and play a critical role in nearly all aspects of biology. Glycans are located on the surfaces of cells and provide cells with the ability to recognize and interact with each other. The expression of carbohydrates helps our immune system to identify viruses and bacteria, determine whether cells are healthy or diseased, and dictates the manner in which the immune system processes these cells, responds to injury, and mediates tissue repair. Glycans are recognized by direct interactions with proteins, often in the context of distinct neighboring peptide motifs. While many drugs target proteins to treat disease, glycans and glycan-protein interactions represent a unique and large class of druggable targets that could be used to develop new treatments. We are developing tools to study the roles of glycan-protein interactions in innate immunity, thrombosis, and cancer and identifying molecules that target these interactions as therapeutic agents.
